Principle of the Assay::The Human PP5 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human PP5 in Cell Culture Supernatants, Serum. This assay employs an antibody specific for Human PP5 coated on a 96-well plate. Standards and samples are pipetted into the wells and PP5 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human PP5 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of PP5 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: TFPI2, also called Tissue factor pathway inhibitor 2, is a protein that in humans is encoded by the TFPI2 gene. It is mapped to 7q21.3. This gene encodes a member of the Kunitz-type serine proteinase inhibitor family. The protein can inhibit a variety of serine proteases including factor VIIa/tissue factor, factor Xa, plasmin, trypsin, chymotryspin and plasma kallikrein. This gene has been identified as a tumor suppressor gene in several types of cancer.
